چکیده

Levels of mRNA for glucokinase, L-pyruvate kinase, fructose-1,6-bisphosphatase and phosphoenolpyruvate carboxykinase were analysed during liver regeneration. Levels of mRNA for glycolytic enzymes (glucokinase and L-pyruvate kinase) decreased rapidly after partial hepatectomy. Glucokinase mRNA increased at 16-24 h, returning to normal values after this time. L-pyruvate kinase mRNA recovered control levels at 168 h. In contrast, phosphoenolpyruvate carboxykinase mRNA increased rapidly after liver resection and remained high during the regenerative process. However, the levels of fructose1,6-bisphosphatase mRNA were not modified significantly. These results correlate with the reported increased rate of gluconeogenesis and changes in enzyme levels after partial hepatectomy. The effect of stress on the mRNA levels was also studied. All enzymes showed variations in their mRNA levels after the surgical stress. In general, the differences were more pronounced in regenerating liver than in sham-operated animals, being practically normalized at 24 h.

Insulin and diet-induced changes in the ubiquitin-modified proteome of rat liver

Ubiquitin is a crucial post-translational modification regulating numerous cellular processes, but its role in metabolic disease is not well characterized. In this study, we identified the in vivo ubiquitin-modified proteome in rat liver and determined changes in this ubiquitome under acute insulin stimulation and high-fat and sucrose diet-induced insulin resistance. We identified 1267 ubiquiti...

Estimation of the Fructose 1,6=Diphosphatase- phosphofructokinase Substrate Cycle and Its Relationship to Gluconeogenesis in Rat Liver in Vito*

Substrate cycling of fructose 6-phosphate through reactions catalyzed by phosphofructokinase and fructose 1,6diphosphatase was estimated in rat liver in vivo. The rate of phosphorylation of fructose 6-phosphate was determined by measuring the release of tritium to intrahepatic water following the metabolism of intraperitoneally injected [5-3H, U-W]glucose. For net flux of hepatic glucose metabo...

Influence of glucagon on the metabolism of xylitol and dihydroxyacetone in the isolated perfused rat liver.

Glucagon stimulates glucose synthesis from xylitol and dihydroxyacetone in the isolated perfused rat liver. In addition to increasing the initial rate of glucose production from these substrates, the hormone also increases the efficiency of gluconeogenesis causing at least 90% of the substrate consumed by the liver to be converted to glucose. These effects of glucagon on gluconeogenesis are obs...
